Overview

Purpose: Orbitrap Fusion Tribrid°ú MSn Library¸¦ ÀÌ¿ëÇÏ¿© MetboliteÀÇ ±¸Á¶¸¦ ¹àÈ÷°íÀÚ ÇÑ´Ù.
Methods: UHPLC¿Í Orbitrap FusionÀ» ¿¬µ¿Çؼ­ Global Profiling°ú MSnÀ» ÀÌ¿ëÇÏ¿© ±¸Á¶ÀûÀÎ ÇØ¼®À» µ¿½Ã¿¡ ÇϰíÀÚ ÇÑ´Ù.
Results: UHPLCÀÇ Timescale(2-3 second FWHM)³»¿¡, 11°³ÀÇ ¼ººÐÀ» Ion Trap HCD MS2¿Í °¢ MS2-MS3(top2)-MS4(top2)ÀÇ Ion Tree µ¥ÀÌÅ͸¦ 10 Cycles Time À̳»·Î ºÐ¼®ÇÒ ¼ö ÀÖ´Ù.

Methods Sample Preparation ¼ºÀÎ ³²¼ºÀ¸·ÎºÎÅÍ ¾òÀº ¼Òº¯ »ùÇÃÀ» ¹°·Î 1/10¹è Èñ¼®Çß°í 3kD MWCO ÇÊÅͷΠó¸®Çß´Ù. Ç÷Àå »ùÇÃÀ» À§Çؼ­, Cold Methanol°ú ¿ø½ÉºÐ¸®±â¸¦ ÀÌ¿ëÇÏ¿© 35°C¿¡¼­ centrifugal evaporation¿¡ ÀÇÇØ ´Ü¹éÁúÀ» Á¦°ÅÇß´Ù. ÀÜ¿©¹°À» MeOH/Water(1/9)·Î ³ì¿©¼­ 3kD MWCO ÇÊÅͷΠó¸®Çß´Ù.
Liquid Chromatography UHPLC ºÐ¸®´Â Hypersil GOLD RP C18¿Í Dionex Ultimate 3000 HPG (High-Pressure Gradient) ÆßÇÁ¸¦ ÀÌ¿ëÇÏ¿© Ä÷³¿Âµµ 55°C ¿¡¼­ 450μL/minÀ¸·Î ºÐ¸®Çß´Ù. LC ¿ë¸Å´Â 0.1% FA (A) °ú 0.1% FA in MeOHÀ¸·Î ¾Æ·¡¿Í °°Àº Á¶°Ç¿¡¼­ ºÐ¸®Çß´Ù.
